The design of graphene-based nanomaterials for biomedical purposes is of great interest. Graphene oxide (GO) nanosheets represent the most widespread type of graphene materials in biological investigations. In the present study, GO nanosheets were synthesized and subsequently chemically functionalized with carboxymethylated dextran (CMD) molecules, referred to as CMD-GO. Then, the effect of molecular weight (MW) and surface concentration (SC) of CMD on the cytotoxicity of the as-prepared CMD-GO were evaluated. Compared with bare GO, CMD-GO nanosheets showed significantly reduced cellular toxicity. Cellular biocompatibility experiments on the effects of MW and SC of CMDs showed that both these parameters, especially the SC, play an important role in the cellular biocompatibility of the synthesized nanohybrids. In fact, the results indicated that the highest level of biocompatibility was related to the CMD-GO with low MW and high SC of CMD. These results offer a previously unavailable nanobiological understanding of to what extend overlooked factors like MW and SC of a polysaccharide coating like CMD can affect the cellular biocompatibility of GO nanosheets.
