Background: H2O2 is used as a vital intermediate for numerous biological reactions, and its detection is attracting attention. Horseradish Peroxidase (HRP) is a brilliant biocatalyst extensively used in several fields of research as well as industry. Aims: Our aim was to prepare a simple, sensitive and cost effective detection of H2O2 becomes quite important for both medical and environmental applications. Methods: One-step GA@MNPs were synthesized using co-precipitation methods. L-lysine functionalization was done via partial oxidation of carbohydrate aldehyde groups. HRP was immobilized on K-GA@MNPs using glutaraldehyde linker. The synthetized nanoparticle was characterized by TEM, FTIR, VSM and XRD analysis. Result and Discussions: The immobilization efficiency was 64.3% and the immobilized HRP retained 60% of its initial activity up to 5 times reusing the enzyme. The pH and temperature optima were shifted from 7.0 for soluble HRP to 8.0 with improved stability from 60 to 70 °C after HRP immobilization, respectively. The synthetized nanostructure exhibited a linear range and detection limitation of 0.1-20 µM, and 0.05 µM, respectively.
