Abstract: Alginate microparticles are considered a very precious tool in pharmaceutical and biomedical applications because of biocompatibility and biodegradability. Microparticles size and shape play a significant role in their applications like drug delivery and cell encapsulation. Here, we report a microfluidic external gelation process using calcium chloride as a crosslinker for producing highly- monodispresed and size-tunable calcium alginate (Ca-alginate) microparticles. The PDMS microfluidic device was fabricated using light lithography procedure and comprised of a cross junction chip with two side channels for mineral oil and a middle channel for alginate solution. When the dispersed phase flow rate was kept constant in 1 ml/min, and the continuous phase flow rates were considered 4, 6, and 8 ml/min, respectively, Alginate droplets with an average diameter ranging from 150 μm to 80 μm were observed. The present experimental results were also compared with the results of numerical simulations, and an acceptable agreement was observed. Matching the experimental results with the numerical results of the formation of alginate nanoparticles can support the theoretical concepts of droplet formation in multiphase medium and precise microchip design.
