The linear globular dendrimer generation 2 (G2/G2-COOH) shows the suitable biocompatibility in the polymeric based delivery systems. Nevertheless, a few studies have been done on the physicochemical properties related to the cellular uptake of these carriers. The surface chemistry has been considered to successful cell internalization. The purpose of this report is to evaluate the cell uptake of the G2 rely on the surface chemistry. In this study, the G2 dendrimers based on the Positively charged (G2-NH2) was synthesized. HEK-293 cells were treated with each type of the G2 dendrimers. The G2 derivatives cytotoxicity and their cellular uptake were evaluated by MTT assay and flow cytometry techniques, respectively. The cytotoxicity results showed that there is no significant difference in the toxicity between G2 and G2-NH2. Nevertheless, the cellular uptake rates of the G2 dendrimers shows the surface chemistry-dependent in the order of G2-NH2> G2-COOH. Increasing in the cellular uptake without changing in the toxicity profile of this dendrimers indicate that the positively charged of these polymers can propose as a suitable strategy for enhancement of the treatments efficiency for them.
