The aim of this study was to evaluate the effect of silk fibroin nanoparticles containing doxorubicin on the expression of p53 gene and its toxicity in-vitro in breast cancer cell line compared to human foreskin fibroblast cells and untreated MCF-7 as normal cells and negative control, respectively. First, sedimentation and separation of silk fibrin proteins were performed using salt and then, protein nanoparticles were prepared using spray-freezing method. Then, the designed nanostructures were characterized by FTIR and DSC. Finally, p53 mRNA levels and cytotoxicity of treated cell lines was evaluated by real time PCR and MTT assay, respectively. Statistical analysis of data and plotting of graphs were performed using Graph Pad Prism. Due to the abnormality of data distribution, Mann-Whitney U test was used to compare the study groups with controls. Characterization data showed that nanostructure synthesis process was well done . Cytotoxicity findings showed a significant inhibition of cell growth in a dose-dependent manner compared to the control group. Also, IC50 values were 0.229 and 0.647 mg/ml for MCF7 and HFF cell line, respectively. There was a significant increase in mRNA level of p53 in MCF-7 cells as well as a significant decrease of p53 gene expression in HFF cells compared to the controls. Doxorubicin-Silk fibroin nanoparticles causes a significant change in cell growth and apoptosis induction.
