: Lung cancer is the first and most common cancer in men and the third most common cancer in women. Nanotechnology applications to diagnose and treat cancers are already a reality providing a wide range of new tools and possibilities. Iron oxide Fe3O4 nanoparticles are used as magnetic data storage, biosensors, drug or siRNA delivery and PEGylation (PEG) has the ability to enhance the retention time of the therapeutics like proteins, enzymes small molecular drugs, liposomes and nanoparticles by protecting them against various degrading mechanisms active inside a tissue or cell, which consequently improves their therapeutic potential. Small interfering RNA (siRNA) is a class of double-stranded RNA non-coding RNA molecules, typically 20-27 base pairs in length, similar to miRNA, and operating within the RNA interference (RNAi) pathway. Since the desired properties of nano-carriers include siRNA protection from degradation and escape from immune recognition in blood, we designed a PEG-coated iron nanocarrier. for the result, we used FT-IR, XRD, DLS, TEM and SEM to identify these nanocarriers. For the biology part, we evaluated the toxicity of this nanocarrier by MTT assay. After that, we used this nanocarrier to target WNT2 gene into SCLC (small cell lung carcinoma) cancer cells and measured gene expression.
