Fluorouracil (5FU) is an antimetabolite drug used in the treatment of various malignancies, such as colon and skin cancers. However, its systemic administration results in severe side effects. Topical 5-FU delivery for the treatment of skin cancer could circumvent these shortcomings, but it is limited by the drug poor permeability through the skin. Here, we introduced a micellar hydrogel to enhance 5FU efficacy against skin cancer and reduce its systemic side effects. 5FU was incorporated into deoxycholic acid (DCA) as a self-assembled micelle. 5FU incorporated DCA micelle was then combined with carboxymethyl chitosan hydrogel (CMC Hyd) to develop a pH-responsive micellar hydrogel system. The properties of the micellar hydrogel system were determined by Fourier-transform infrared spectroscopy (FT-IR), Dynamic light scattering (DLS), zeta potential, and atomic force microscopy (AFM). Drug release in aqueous media shows pH-dependent behavior. The 5FU@Mic-Hyd system can be implemented in developing pH-triggered delivery systems to achieve the desired release profile.
