Type 1 diabetes mellitus (T1DM) is a global concern that affects a large population worldwide. The standard treatment for this disease was still unable to cure some crucial complications. The present study aimed to evaluate a microfluidic chip's potency to encapsulate the trabecular meshwork mesenchymal (TMMSCs) in alginate microgel and differentiate them into Insulin Producing Cells (IPCs). TMMSCs were isolated and proliferate to the suitable confluency. A microfluidic channel chip was employed to encapsulate TMMSCs in alginate microgel. MTT assay and RT-qPCR were used to investigate TMMSCs viability and IPC differentiation, respectively. RT-qPCR analysis has shown that Insulin and PDX-1 genes have significantly increased in 3D microgels encapsulated cells compared to TCPS. Finally, our data showed the potency of microgel to support the encapsulated TMMSCs differentiation. Keywords: Microfluidic, Microchip, MSCs, Stem cell, Tissue engineering
